Technology: The Inventions That Make Us Go Wow!

Discovering the Cut-and-Paste Enzymes A diagram shows a circular grey plasmid in column 1 and a circular red plasmid in column 2. Both plasmids look like two concentric cir-cles; each circle represents a DNA strand. Different restriction en-zymes are used to cleave, or cut, plasmid 1 and plasmid 2. \"Sticky ends,\" or overhanging DNA ends without a complementary strand, are treated with an enzyme that digests single-stranded DNA. New complementary sticky ends are then added by terminal transferase. dATP is added to one plasmid, and dTTP is added to the other plas-mid to produce poly-A and poly-T sticky ends, respectively. After the addition of complementary sticky ends to plasmids 1 and 2, the two plasmids are mixed together, and the complementary sticky ends base pair. A recombined plasmid is shown in a single, center col-umn. The recombined plasmid is composed of two larger concentric circles; half of the circle is grey, and the other half is red. DNA poly-merase, shown as a blue enzyme encircling both DNA strands, is added to the new, recombined plasmid to insert missing nucleo-tides. DNA ligase, shown as a small yellow enzyme encircling one DNA strand, seals nicks in the sugar-phosphate groups to ensure the fragments from each plasmid are joined together. A second major step forward in gene modification was the discovery of restriction enzymes, which cleave DNA at specific sequences. These enzymes were discovered at approximately the same time as the first DNA ligases by Swiss biologist Werner Arber and his col-leagues while they were investigating a phenomenon called host-controlled restriction of bacteriophages. Bacteriophages are viruses that invade and often destroy their bacterial host cells; host-controlled restriction refers to the defense mechanisms that bacteri-al cells have evolved to deal with these invading viruses. Arber's team discovered that one such mechanism is enzymatic activity pro-vided by the host cell. The team named the responsible enzymes "restriction enzymes" because of the way they restrict the growth of bacteriophages. These scientists were also the first to demonstrate that restriction enzymes damage invading bacteriophages by cleav-ing the phage DNA at very specific nucleotide sequences (now known as restriction sites). View Full-Size ImageFigure 2 The first major step forward in the ability to chemically modify genes occurred when American biologist Martin Gellert and his colleagues from the National Institutes of Health purified and characterized an enzyme in Escherichia coli responsible for the actual joining, or re-combining, of separate pieces of DNA (Zimmerman et al., 1967). They called their find "DNA-joining enzyme," and this enzyme is now known as DNA ligase. All living cells use some version of DNA ligase to "glue together" short strands of DNA during replication. Using E. coli extract, the researchers next showed that only in the presence of ligase was it possible to repair single-stranded breaks in λ phage DNA. (Discovered in 1950 by American microbiologist Esther Leder-berg, λ phage is a virus particle that infects E. coli.) More specifical-ly, they showed that the enzyme was able to form a 3'-5'-phosphodiester bond between the 5'-phosphate end of the last nu-cleotide on one DNA fragment and the 3'-OH end of the last nucleo-tide on an adjacent fragment. The identification of DNA ligase was the first of several key steps that would eventually empower scien-tists to attempt their own recombination experiments—experiments that involved not just recombining the DNA of a single individual, but recombining DNA from different individuals, including different spe-cies.

AT HOME (UP TO 1 DAY AHEAD) 1. Cook potatoes in a large pot of boiling water until larger chunks are tender when pierced with a fork. Drain; return to pot and keep there for transport. Pour half-and-half, salt, and pepper into a jar with tight-fitting lid. Put leek, shal-lot, and green onions in separate resealable plastic bags. Chill all. AT THE PARTY 2. Heat potatoes over low heat, stirring often, just until warm. Meanwhile, heat half-and-half mixture in a microwave until very hot. Add to potatoes and heat, stirring occasionally, over medium heat until hot. Using a hand mixer (or put potatoes in a stand mixer and work in batches if necessary), whip potatoes just until smooth and free of lumps (do not overwhip). Transfer potatoes to a serving bowl. 3. Melt butter in a large frying pan over medium heat. Add leek and cook just until starting to soften, about 3 minutes. Add shallot; cook until softened and butter starts to brown, about 3 minutes. Remove from heat, add green onions, and swirl pan to coat. Pour over potatoes. Ingredients 6 large russet potatoes (about 5 lbs.), peeled and cut into chunks 1 pt. half-and-half 4 teaspoons kosher salt 1/2 teaspoon of pepper 1 leek, white and light green parts only, sliced 1 large shallot, sliced 2 green onions sliced 6 tablespoons of butter

Suicide bomber kills 45 at Afghanistan volleyball game

At least 45 people were killed and 60 others wounded Sunday when a suicide bomber attacked a crowd watching a volleyball match in the Yahyakhil district of Afghanistan's southeastern Paktika province, according to a spokesman for the provincial governor. In 2010, a suicide car bomb exploded in the middle of a group of men playing volleyball, a popular sport in the region, in northwest Pakistan. That attack left 30 people dead and 52 wounded. Sunday's attack comes the same day the nation's parliament agreed to allow U.S. and NATO forc-es to remain in Afghanistan after 2014. It's unusual for an attack in Afghanistan to kill so many people.

Multiple award-winning director Mike Nichols dies at 83 Mike Nichols, the award-winning director and pioneering comedian who was one of the few people to win an Emmy, a Grammy, an Oscar and a Tony award, died Wednesday evening, according to a note from ABC News' president to its staff. Nichols, 83, who directed "Who's Afraid of Virginia Woolf" and "The Graduate," was the husband of former "ABC World News" anchor Diane Sawyer. Nichols' influence looms large in entertainment. He was among the founders of the Compass Players, the predecessor to the Chicago-based comedy troupe Second City, which has been a proving ground for comic talent for more than four decades.

Report: Janice Dickinson accuses Bill Cosby of rape Former supermodel Janice Dickinson accused Bill Cosby of sexual assault, telling "Entertainment Tonight" in an interview aired on Tuesday that the comedian raped her. Dickinson is the latest of a handful of women who have accused Cosby of sexual as-sault -- allegations the comedian has repeatedly said are untrue. He has never been prosecuted. My girlfriend has genital herpes

A girl I've been seeing for some time just told me after already hav-ing sex with her that she has genital herpes. She told me that she had a minor outbreak 6 months ago (1 sore) and that is when she be-gan taking Valtrex. We used a condom but I went down on her. I've read that the statistics of spreading genital herpes while on Valtrex using a condom is 1% but what are the statistics of me going down on her (on Valtrex) obviously unprotected. Any advice is appreciated. How can I protect myself from genital herpes if we keep having sex? While no prevention method short of abstinence is 100% effective, using a latex condom offers some protection. Your partner should tell you when symptoms flare up, which is when the virus is most contagious. Avoid sex when your partner has symptoms. How can I find out if I've been infected with genital herpes? Go to your doctor and get tested. A doctor may take a sample from what appears to be a genital herpes sore and send it to a lab for test-ing. You can also have a blood test. The blood test looks for antibodies to the virus that the immune system would have made when you were infected. The second type of herpes simplex virus, HSV-2, al-most always infects the genitals, so if antibodies to HSV-2 are de-tected in the blood, you probably have genital herpes. A blood test that shows antibodies to the other type of herpes virus, HSV-1, means you could have genital or oral herpes. That's because oral herpes, typically caused by HSV-1, can be spread to the genitals

dur-ing oral sex.

Morning yoga also gives me that calm, quiet time alone before my baby or husband wakes up; I can focus on myself and reflect on all the won-derful things in my life. When I'm done with the poses, I'm in a brighter mood, without that "I just rolled out of bed" grogginess. Really, even a cup of coffee doesn't give you as much long-lasting energy— and java can't firm up your body, either! Step 1 Start standing, hands in prayer at heart center. Close eyes if you’d like and fo-cus on your breath. Step 2 Take 3 to 5 breaths, then inhale and lift hands above head; look up to hands. Step 3 On an exhale, open arms out to sides and swan- dive to the floor, folding into legs. Step 4 Inhale and step right leg back to Crescent Lunge pose. Letting chest lift, float arms above head. Press hands in prayer. Look up to your hands Step 5 Exhale onto hands and knees, then inhale into a swayback Cow pose. Step 6 Tuck toes under and exhale into Downward Dog. Hold for 5 breaths. Step 7 Step right foot forward to Crescent Lunge, then bring left foot to meet right in Standing Forward Bend. Inhale arms back overhead and exhale hands to heart. Align your knee over your ankle.

North Korea digging tunnels in South

Gen. Hahn Sung-Chu never believed North Korea could dig a tunnel that reached Seoul -- until now.

Standing inside a basement of an apartment block in the heart of the capital, the for-mer two-star general in the South Korean military says, "This is a kind of invasion, North Korean soldiers working underneath us."

Hahn says residents had complained of underground vibrations, but the subway does not run beneath them.

He says dowsers detected three tunnels, 13 to 16 feet (4 to 5 meters) wide at a depth of up to 39 feet (12 meters). His team drilled two bore holes to lower a camera, but before they could, they detected two underground explosions and their drill holes were blocked. Hahn is certain that North Korean soldiers were working beneath their feet, protecting the tunnel.

Secret Service Director Julia Pierson resigns

Julia Pierson, the first female director of the Secret Service, resigned Wednesday in the aftermath of a fence-jumper gaining access to the White House on September 19 and a subsequent congressional inquiry uncovering other security lapses.

Homeland Security Director Jeh Johnson announced the resignation in a statement. He also announced that the Department of Homeland Security would take over an in-ternal inquiry of the Secret Service and that he would appoint of a new panel to re-view security at the White House.

Joseph Clancy, formerly a special agent in charge of the Presidential Protective Divi-sion of the Secret Service, was named interim director, Johnson said in his statement.

Researchers develop new cells meant to form blood vessels, treat peripheral artery disease

Researchers have developed a technique to jump-start the body's systems for creating blood vessels, opening the door for potential new treatments for dis-eases whose impacts include amputation and blindness.

The international team, led by scientists at the Indiana University School of Med-icine, is targeting new therapies for illnesses such as peripheral artery disease, a painful leg condition caused by poor blood circulation. The disease can lead to skin problems, gangrene and sometimes amputation.

While the body has cells that specialize in repairing blood vessels and creating new ones, called endothelial colony-forming cells, these cells can lose their abil-ity to proliferate into new blood vessels as patients age or develop diseases like peripheral arterial disease, said Mervin C. Yoder Jr., M.D., Richard and Pauline Klingler Professor of Pediatrics at IU and leader of the research team.

Peripheral artery disease patients can be given medication to improve blood flow, but if the blood vessels to carry that improved flow are reduced in number or function, the benefits are minimal. If "younger," more "enthusiastic" endothe-lial colony forming cells could be injected into the affected tissues, they might jump-start the process of creating new blood vessels. Gathering those cells would not be easy however -- they are relatively difficult to find in adults, espe-cially in those with peripheral arterial disease. However, they are present in large numbers in umbilical cord blood.

Reporting their work in the journal Nature Biotechnology, the researchers said they had developed a potential therapy through the use of patient-specific in-duced pluripotent stem cells, which are normal adult cells that have been "coaxed" via laboratory techniques into reverting into the more primitive stem cells that can produce most types of bodily tissue. So, in one of the significant discoveries reported in the Nature Biotechnology paper, the research team de-veloped a novel methodology to mature the induced pluripotent stem cells into cells with the characteristics of the endothelial colony-forming cells that are found in umbilical cord blood. Those laboratory-created endothelial colony-forming cells were injected into mice, where they were able to proliferate into human blood vessels and restore blood flow to damaged tissues in mouse reti-nas and limbs.

Scientists watch proteins move on surface of HIV virus in real-time

The research team, including co-lead author Dr. Scott Blanchard of Weill Cornell Medical College in New York, NY, says the discovery may shed light on how the HIV virus infects human immune cells, paving the way for strategies that prevent such infection.

"Making the movements of HIV visible so that we can follow, in real-time, how surface proteins on the virus behave will hopefully tell us what we need to know to prevent fusion with human cells - if you can prevent viral entry of HIV into im-mune cells, you have won," explains Dr. Blanchard.

In their study, Dr. Blanchard and colleagues describe how they created fluores-cent molecules, referred to as "beacons," and introduced them to the outer cov-ering of the HIV virus - called the "envelope."

They then modified a technique called single-molecule fluorescence resonance energy transfer (smFRET) imaging to watch two of the beacons.

This imaging method uses fluorescent light to measure the distance viral parti-cles travel. In this study, the team were able to measure distances - down to a billionth of an inch - between the two beacons, which lit-up in different colors. The team were then able to detect real-time shape-shifting of the virus as the two beacons moved.

Using this technology, the researchers were able to analyze the movements of envelope proteins - gp120 and gp41, known as "trimers" - on the surface of the HIV virus.

Such proteins are important in allowing the HIV virus to infect human cells that carry CD4 receptor proteins - the proteins that assist HIV in binding to a cell. The researchers explain that these proteins "open up like a flower" when CD4 is present, revealing a gp41 subunit.

Smoking Finding ways to quit

The findings of the study, published in JAMA Internal Medicine, sug-gest that the disease burden of cigarette smoking in the US "remains immense," write the authors.

Cigarette smoking is acknowledged as the leading cause of prevent-able deaths in the US, with experts finding cigarette smoke capable of harming almost every organ and organ system within the body.

The Centers for Disease Control and Prevention (CDC) regularly publish their estimates of mortality attributable to smoking, along with its economic cost. However, the population disease burden tends to receive less focus, despite smoking's status as a leading cause for many major medical conditions.

In 2000, the CDC published estimates of smoking-attributable mor-tality and reported that 8.6 million individuals suffered from a total of 12.7 million medical conditions attributable to smoking.

"Most of these conditions were chronic bronchitis and emphysema, often classified as chronic obstructive pulmonary disease (COPD)," write the authors, "but these estimates and methods, to our knowledge, have not been subsequently updated or refined."

Thyroid Disease

Last year, while sitting through a medical board review course for integrative medicine, an inexplicable feeling of fatigue washed over me. I had been sleeping well, yet still continuing to wake up exhausted. It was no small coincidence that the topic of discussion was the scientific evidence behind the integrative medicine approach to treating thyroid disease. You needn't have a medical background to know my symptoms were not normal. My hairdresser was watching my hair fall out by the handfuls for months, and she kept asking me about potential thyroid issues.

Have you also suffered from similar symptoms that could be related to thyroid disease? Subclinical thyroid disease and hypothyroidism are both more common in women than in men in the United States. I had gone to my primary care physician many times over the years complaining of symptoms. My thyroid stimulating hormone (TSH) levels always came back in the upper limit of normal range (around 10 mIU/L) so therefore no other tests were done. This is the problem. Often, to screen potential thyroid issues, physicians only check this one hormone level, which is created in the brain.

The serum TSH has a wide range of normal (from 4-10 IU/L), and your own TSH level's range of normal may be less than a calculated average for the entire population. According to the 2012 American Association of Clinical Endocrinology Guidelines, there are many reasons for a varying TSH level. Checking a serum TSH level is a screening test only, and not truly designed to diagnose a thyroid problem or monitor therapy. These guidelines recommend that if a patient is symptomatic, the additional two main thyroid hormones should also be checked: thyroxine (T4) and triidoothyronine (T3) or FT4 and FT3 levels.
