Technology: The Inventions That Make Us Go Wow!

Discovering the Cut-and-Paste Enzymes  A diagram shows a circular grey plasmid in column 1 and a circular red plasmid in column 2. Both plasmids look like two concentric cir-cles; each circle represents a DNA strand. Different restriction en-zymes are used to cleave, or cut, plasmid 1 and plasmid 2. \"Sticky ends,\" or overhanging DNA ends without a complementary strand, are treated with an enzyme that digests single-stranded DNA. New complementary sticky ends are then added by terminal transferase. dATP is added to one plasmid, and dTTP is added to the other plas-mid to produce poly-A and poly-T sticky ends, respectively. After the addition of complementary sticky ends to plasmids 1 and 2, the two plasmids are mixed together, and the complementary sticky ends base pair. A recombined plasmid is shown in a single, center col-umn. The recombined plasmid is composed of two larger concentric circles; half of the circle is grey, and the other half is red. DNA poly-merase, shown as a blue enzyme encircling both DNA strands, is added to the new, recombined plasmid to insert missing nucleo-tides. DNA ligase, shown as a small yellow enzyme encircling one DNA strand, seals nicks in the sugar-phosphate groups to ensure the fragments from each plasmid are joined together.  A second major step forward in gene modification was the discovery of restriction enzymes, which cleave DNA at specific sequences. These enzymes were discovered at approximately the same time as the first DNA ligases by Swiss biologist Werner Arber and his col-leagues while they were investigating a phenomenon called host-controlled restriction of bacteriophages. Bacteriophages are viruses that invade and often destroy their bacterial host cells; host-controlled restriction refers to the defense mechanisms that bacteri-al cells have evolved to deal with these invading viruses. Arber's team discovered that one such mechanism is enzymatic activity pro-vided by the host cell. The team named the responsible enzymes "restriction enzymes" because of the way they restrict the growth of bacteriophages. These scientists were also the first to demonstrate that restriction enzymes damage invading bacteriophages by cleav-ing the phage DNA at very specific nucleotide sequences (now known as restriction sites). The identification and characterization of

View Full-Size ImageFigure 2 The first major step forward in the ability to chemically modify genes occurred when American biologist Martin Gellert and his colleagues from the National Institutes of Health purified and characterized an enzyme in Escherichia coli responsible for the actual joining, or re-combining, of separate pieces of DNA (Zimmerman et al., 1967). They called their find "DNA-joining enzyme," and this enzyme is now known as DNA ligase. All living cells use some version of DNA ligase to "glue together" short strands of DNA during replication.  Using E. coli extract, the researchers next showed that only in the presence of ligase was it possible to repair single-stranded breaks in λ phage DNA. (Discovered in 1950 by American microbiologist Esther Leder-berg, λ phage is a virus particle that infects E. coli.) More specifical-ly, they showed that the enzyme was able to form a 3'-5'-phosphodiester bond between the 5'-phosphate end of the last nu-cleotide on one DNA fragment and the 3'-OH end of the last nucleo-tide on an adjacent fragment. The identification of DNA ligase was the first of several key steps that would eventually empower scien-tists to attempt their own recombination experiments—experiments that involved not just recombining the DNA of a single individual, but recombining DNA from different individuals, including different spe-cies.

Cleveland police say slain youth held air gun 'indistinguishable from a real gun'

The 12-year-old boy shot by Cleveland police over the weekend was wielding an air gun that is "indistinguishable from a real gun," Cleveland Police Chief Calvin Williams told reporters Monday. [Previous story, posted at 10:41 a.m. ET] Cleveland authorities have surveillance video showing the weekend shooting by po-lice of a 12-year-old boy who was brandishing an ultrarealistic air gun, a department spokesman said Monday.

Exclusive: Why TV anchors met secretly with Ferguson officer who shot Michael Brown

In the pursuit of that interview, several high-profile television anchors have secretly met with Wilson, according to sources at several TV networks. All of the meetings were off the record, meaning the anchors could not describe what was said. These meetings are a normal part of the TV guest booking process, but they're signifi-cant in this case because Wilson has not been seen in public since Brown's death in Ferguson, Missouri on August 9. Among the anchors who have met with Wilson are Matt Lauer of NBC, George Steph-anopoulos of ABC, Scott Pelley of CBS, and Anderson Cooper and Don Lemon of CNN.

Suicide bomber kills 45 at Afghanistan volleyball game

At least 45 people were killed and 60 others wounded Sunday when a suicide bomber attacked a crowd watching a volleyball match in the Yahyakhil district of Afghanistan's southeastern Paktika province, according to a spokesman for the provincial governor. In 2010, a suicide car bomb exploded in the middle of a group of men playing volleyball, a popular sport in the region, in northwest Pakistan. That attack left 30 people dead and 52 wounded. Sunday's attack comes the same day the nation's parliament agreed to allow U.S. and NATO forc-es to remain in Afghanistan after 2014. It's unusual for an attack in Afghanistan to kill so many people.

Wall Street Leading Washington Yet Again: What Was Obama Thinking?

If you want to understand what makes Elizabeth Warren so special in American poli-tics, consider her nervy leadership of the campaign to block President Obama's foolish nomination of one Antonio Weiss to be the top Treasury official in charge of the do-mestic financial system, including enforcement of the Dodd-Frank Act. For most of his Wall Street career, Weiss has epitomized everything that reeks about financial abuses. As chief of international mergers and acquisitions for Lazard, Weiss orchestrated what are delicately known as "corporate inversions," in which a domestic corporation moves its nominal headquarters offshore, to avoid its U.S. taxes. It's hard to improve on Sen. Warren's description of this play, in her Huffington Post blog of last Wednesday:

Iran Nuclear Talks To Be Extended After Missing Deadline VIENNA, Nov 24 (Reuters)

 - Iran and six powers failed for a second time this year on Monday to resolve their 12-year dispute over Tehran's nuclear ambitions and gave themselves seven more months to overcome the deadlock that has prevented them from clinching an historic deal. Western officials said they were aiming to secure an agreement on the substance of a final accord by March but that more time would be needed to reach a consensus on the all-important technical details. "We have had to conclude it is not possible to get to an agreement by the deadline that was set for today and therefore we will extend the JPOA to June 30, 2015," British For-eign Secretary Philip Hammond told reporters at the end of the talks.

What's Wrong With This Picture? For U.S. Fight Against ISIS, Everything WASHINGTON

-- The Islamic Republic of Iran would like to make one thing clear: We've got this. Up until June 10, Iranian officials had been content to shape events in Iraq quietly through their hold on local Shiite militias and the prime minister at the time, Nouri al-Maliki. Then the Iraqi government lost Mosul, the nation's second-largest city, to the growing Sunni extremist force now known as the Islamic State, or ISIS. The U.S. eventually re-sponded: It forced a new government to take power, sent in airpower and military ad-visers, and launched an international effort against ISIS in both Iraq and Syria. But months before the U.S. showed that it was willing to invest heavily in the region again, Iran decided the rise of ISIS gave it the chance to stop being coy about its con-trol of the Iraqi government.

During NCAA review, UNC gets advice from former infractions chief There are few professors as involved in NCAA matters as University of Nebraska law professor Jo Potuto. For more than a decade, she has served on the panel that hands down punishments to universities that violate NCAA regulations, including two years as its chair. She has helped shape those regulations on NCAA committees, in opinion pieces, lectures and congressional testimony. She’s also one of the nation’s longest-serving faculty athletic representatives, holding that role at Nebraska for 17 years. She receives no pay for her NCAA work. But several weeks ago, she began a paid consulting job assisting UNC-Chapel Hill with what appears to be a landmark case of academic fraud that benefited athletes and is now under NCAA investigation.   Venerable Durham Arts Council celebrates 60 years amid buzz of new scene New art studios and upstart galleries have generated a trendy buzz about the arts belt stretching through downtown Durham. But it was one of the city’s mainstays – the venerable Durham Arts Council building – that was creating the hum during the weekend. Fine artists, jewelry makers, fashion designers, ceramicists and more took part in a two-day art walk and holiday market that was as much about supporting the arts, in general, as it was the artists individually.  Moms, daughters stuff homemade stockings for military families Mothers and daughters sprang into action Sunday at the Embassy Suites, stuffing col-orful felt Christmas stockings to the brim with toys, candy and toothbrushes for chil-dren of service members. Even Santa was there, mingling and taking photos with his part-time elves from the Carolina Lily Chapter of the National Charity League Inc. “This is exciting. They did a lot of work putting all the stockings together,” said Diane D’Agostine, who made 50 stockings with her daughters Paige and Devin. “The best part is going to the USO when they do their Holiday in the Hangar party, and you get to hand them out.”   The nonprofit National Charity League works with girls in grades seven through 12 and their mothers, offering diverse ways to help communities and build leadership skills.  This place is really good. Yes, just like many of you, we walked by this giant space when it was being built and thought, "why is this massive place going in on 2nd street?" and "another taco spot? don't we have enough on this strip?" My husband and I just couldn't imagine that this place would be a good fit at all, nor was it necessary. But we always like to support our local restaurants, so we thought, let's give it a try, and then we'd really have some ammunition for our unfounded hatred.

My girlfriend has genital herpes

A girl I've been seeing for some time just told me after already hav-ing sex with her that she has genital herpes. She told me that she had a minor outbreak 6 months ago (1 sore) and that is when she be-gan taking Valtrex. We used a condom but I went down on her. I've read that the statistics of spreading genital herpes while on Valtrex using a condom is 1% but what are the statistics of me going down on her (on Valtrex) obviously unprotected. Any advice is appreciated.  How can I protect myself from genital herpes if we keep having sex?  While no prevention method short of abstinence is 100% effective, using a latex condom offers some protection. Your partner should tell you when symptoms flare up, which is when the virus is most contagious. Avoid sex when your partner has symptoms.  How can I find out if I've been infected with genital herpes?  Go to your doctor and get tested. A doctor may take a sample from what appears to be a genital herpes sore and send it to a lab for test-ing.  You can also have a blood test. The blood test looks for antibodies to the virus that the immune system would have made when you were infected. The second type of herpes simplex virus, HSV-2, al-most always infects the genitals, so if antibodies to HSV-2 are de-tected in the blood, you probably have genital herpes. A blood test that shows antibodies to the other type of herpes virus, HSV-1, means you could have genital or oral herpes. That's because oral herpes, typically caused by HSV-1, can be spread to the genitals

dur-ing oral sex.

Yoga: Natural Healing and body connections

Good morning yoga  I love to wake up and greet the day with a Sun Salutation (or Surya Namaskar, in Sanskrit). Not only does it let me acknowledge yet another beautiful morning, but I always feel so much better afterward. It's a chance to stretch out all the kinks from the night and connect with my breath so I can set my intention—or gal— for the day. Morning yoga also gives me that calm, quiet time alone before my baby or husband wakes up; I can focus on myself and reflect on all the won-derful things in my life. When I'm done with the poses, I'm in a brighter mood, without that "I just rolled out of bed" grogginess. Really, even a cup of coffee doesn't give you as much long-lasting energy— and java can't firm up your body, either!   Step 1 Start standing, hands in prayer at heart center. Close eyes if you’d like and fo-cus on your breath.  Step 2 Take 3 to 5 breaths, then inhale and lift hands above head; look up to hands.  Step 3 On an exhale, open arms out to sides and swan- dive to the floor, folding into legs.  Step 4 Inhale and step right leg back to Crescent Lunge pose. Letting chest lift, float arms above head. Press hands in prayer. Look up to your hands  Step 5 Exhale onto hands and knees, then inhale into a swayback Cow pose.  Step 6 Tuck toes under and exhale into Downward Dog. Hold for 5 breaths.  Step 7 Step right foot forward to Crescent Lunge, then bring left foot to meet right in Standing Forward Bend. Inhale arms back overhead and exhale hands to heart. Align your knee over your ankle.

Distortion in the Digital Era: New age Technology

The Digitized Economy is shifting value away from established players unable to evolve quickly enough. The speed and size of this shift is tremendous, fueled by yet another wave of new technology powerhouses — the young “unicorn” com-panies wielding billion-dollar valuations to shape entirely new markets, with a glut of private capital lubricating their engines. Such exponential super-startups are even taking the Internet giants by surprise. The disruptors are now facing their own disruption.  According to Sources in “ Meet your Boss” We all know the story about algorithms and work the past few years. Service jobs across the country are increasingly being managed with the help of mathe-matical models of customer demand, revolutionizing everything from taxi driv-ing to food delivery, home cleaning, and laundromats. I have argued that the in-creased autonomy and flexibility of these jobs means that algorithms are taking over unions as the primary driver of workers’ rights in the 21st century.  But now, startups are starting to move up the corporate ladder, using algorithms to improve and disrupt professions that up until recently have seemed almost completely insulated from the efficiencies of computation.  Two Boston-based startups, HourlyNerd and Quantopian, are using sophisticat-ed algorithms to compete with some of the most-well known firms in consulting and quantitative finance, respectively. As they begin to disrupt these two indus-tries, both hope to expand and democratize their professions to anyone with the talent and desire to work.  To help people get a hold of this rapidly changing landscape, I highlighted seven tech-nologies that will help you innovate faster, beat your competition and deliver outstand-ing customer service. They each address the common challenges of how businesses operate today, and are setting the stage for a new era of IT. ElasticBox Zimperium ZenPayroll OneLogin Conjur Illumio RelateIQ

Dementia With Lewy Bodies

Dementia is a progressive (gradually worsening) decline of mental abilities that disturbs "cognitive" functions such as memory, thought processes, and speech as well as behavior and movements.  Dementia with Lewy bodies (DLB) is the name for a group of disorders in which dementia is caused by the presence of Lewy bodies in the brain. Lewy bodies are small round clumps of normal proteins that -- for unknown reasons -- be-come abnormally clumped together inside neurons (brain cells). Whether the Lewy bodies directly cause gradual damage to the brain cells, impairing their function and eventually killing them, or are only a marker of some other de-structive process, is not known.  Lewy bodies are named after Frederic Lewy, the doctor who first described them in 1912.  Lewy first found Lewy bodies in the brains of people with Parkin-son's disease. Parkinson's disease is a condition best known for disrupting body movements. The most common of these "motor" symptoms are tremor (shaking or trembling) of the hands (that mainly occurs when the hands are at rest and not moving), rigidity (stiffness) of the trunk and limbs, slowness of movement, and loss of balance and coordination. An estimated 30% to 60% of people with Parkinson's disease develop dementia.   Scientists later discovered cases of Alzheimer's-type dementia linked to Lewy bodies. This was thought to be very rare, but as tests of brain tissue improved, it became clear that Lewy bodies were fairly common and were linked to several different types of dementia. A type of dementia similar to but different from Alz-heimer's disease was recognized and called dementia with Lewy bodies, or DLB. DLB is now believed to be the second or third most common type of de-mentia after Alzheimer's disease, accounting for about 10% to 20% of all de-mentias. (There is controversy about whether DLB or vascular dementia is the second most common form of dementia.)

Aaron Hernandez case: Who's who?

Former New England Patriot Aaron Hernandez is behind bars, accused of murdering three people.

Long before he made national headlines, he was a standout athlete in Bristol, Connect-icut, who came from a family described as a local sports dynasty. Hernandez left high school halfway through his senior year in January 2007 to join the University of Florida Gators. He entered the NFL draft after his junior year and was selected by the Patriots in the fourth round.

By the end of the second season, he inked a five-year contract extension worth $40 million. But less than a year later, he was charged with first-degree murder in the death of Odin Lloyd. Two hours after his arrest on June 26, 2013, the Patriots dropped him from the team.

Hernandez has also been charged in the slayings of Daniel de Abreu and Safiro Furta-do, and some of his closest

North Korea digging tunnels in South

Gen. Hahn Sung-Chu never believed North Korea could dig a tunnel that reached Seoul -- until now.

Standing inside a basement of an apartment block in the heart of the capital, the for-mer two-star general in the South Korean military says, "This is a kind of invasion, North Korean soldiers working underneath us."

Hahn says residents had complained of underground vibrations, but the subway does not run beneath them.

He says dowsers detected three tunnels, 13 to 16 feet (4 to 5 meters) wide at a depth of up to 39 feet (12 meters). His team drilled two bore holes to lower a camera, but before they could, they detected two underground explosions and their drill holes were blocked. Hahn is certain that North Korean soldiers were working beneath their feet, protecting the tunnel.

Secret Service Director Julia Pierson resigns

Julia Pierson, the first female director of the Secret Service, resigned Wednesday in the aftermath of a fence-jumper gaining access to the White House on September 19 and a subsequent congressional inquiry uncovering other security lapses.

Homeland Security Director Jeh Johnson announced the resignation in a statement. He also announced that the Department of Homeland Security would take over an in-ternal inquiry of the Secret Service and that he would appoint of a new panel to re-view security at the White House.

Joseph Clancy, formerly a special agent in charge of the Presidential Protective Divi-sion of the Secret Service, was named interim director, Johnson said in his statement.

Trendy News: Oprah

Oprah Winfrey

Is Shopping with bff Gail King spending more then 2.9million dollars. Putting a small economic damp on our economy for reasons of nothing but enjoying a day of shopping. Another scandal upon which we can say some-thing as controversial as lying to Harvard. Lets remind our self's again what happened. According to other sources, here is something special about the optimism of the very privileged. At Harvard not long ago, Oprah Winfrey spoke to the graduating class and, "in] my remarks to anybody who has ever felt inferi-or or felt disadvantaged, felt screwed by life," uttered this memorable sentence: "There is no such thing as failure." She immediately explained her strange assertion: "Failure is just life trying to move us in another direction." The experience of defeat, in other words, is an error of interpretation. Nothing is bad that is followed by something else. Nothing is bad unless you call it bad. Only death, on this ac-count, is a defeat, since it is followed by nothing, though I suppose that in her quackery Winfrey be-lieves in an afterlife in which she dwells for eternity between Tom Cruise and Maya Angelou at God’s Oscar party. ….From Our sources at Harvard….. But we do like the flashback and reminders of what looking beyond a little white lie can do for everyone's sake.

Gadget Ogling: Jump Starts, Wearable Drones, Smarter Yoga and Safer Food

Welcome, dear readers, to a new installment of Gadget Dreams and Nightmares, a fresh peek at gadgets hurtling down the pipeline toward our homes.

On the docket this time around are a motivational wearable device, a smart yoga mat, a golfer-focused smartwatch, a drone with a difference, and a way to stop food from going bad.

As always, these aren't reviews -- partly because some of these items are nowhere near market-ready yet -- and the ratings denote only my interest in actually using them.

Researchers develop new cells meant to form blood vessels, treat peripheral artery disease

Researchers have developed a technique to jump-start the body's systems for creating blood vessels, opening the door for potential new treatments for dis-eases whose impacts include amputation and blindness.

The international team, led by scientists at the Indiana University School of Med-icine, is targeting new therapies for illnesses such as peripheral artery disease, a painful leg condition caused by poor blood circulation. The disease can lead to skin problems, gangrene and sometimes amputation.

While the body has cells that specialize in repairing blood vessels and creating new ones, called endothelial colony-forming cells, these cells can lose their abil-ity to proliferate into new blood vessels as patients age or develop diseases like peripheral arterial disease, said Mervin C. Yoder Jr., M.D., Richard and Pauline Klingler Professor of Pediatrics at IU and leader of the research team.

Peripheral artery disease patients can be given medication to improve blood flow, but if the blood vessels to carry that improved flow are reduced in number or function, the benefits are minimal. If "younger," more "enthusiastic" endothe-lial colony forming cells could be injected into the affected tissues, they might jump-start the process of creating new blood vessels. Gathering those cells would not be easy however -- they are relatively difficult to find in adults, espe-cially in those with peripheral arterial disease. However, they are present in large numbers in umbilical cord blood.

Reporting their work in the journal Nature Biotechnology, the researchers said they had developed a potential therapy through the use of patient-specific in-duced pluripotent stem cells, which are normal adult cells that have been "coaxed" via laboratory techniques into reverting into the more primitive stem cells that can produce most types of bodily tissue. So, in one of the significant discoveries reported in the Nature Biotechnology paper, the research team de-veloped a novel methodology to mature the induced pluripotent stem cells into cells with the characteristics of the endothelial colony-forming cells that are found in umbilical cord blood. Those laboratory-created endothelial colony-forming cells were injected into mice, where they were able to proliferate into human blood vessels and restore blood flow to damaged tissues in mouse reti-nas and limbs.

Scientists watch proteins move on surface of HIV virus in real-time

The research team, including co-lead author Dr. Scott Blanchard of Weill Cornell Medical College in New York, NY, says the discovery may shed light on how the HIV virus infects human immune cells, paving the way for strategies that prevent such infection.

"Making the movements of HIV visible so that we can follow, in real-time, how surface proteins on the virus behave will hopefully tell us what we need to know to prevent fusion with human cells - if you can prevent viral entry of HIV into im-mune cells, you have won," explains Dr. Blanchard.

In their study, Dr. Blanchard and colleagues describe how they created fluores-cent molecules, referred to as "beacons," and introduced them to the outer cov-ering of the HIV virus - called the "envelope."

They then modified a technique called single-molecule fluorescence resonance energy transfer (smFRET) imaging to watch two of the beacons.

This imaging method uses fluorescent light to measure the distance viral parti-cles travel. In this study, the team were able to measure distances - down to a billionth of an inch - between the two beacons, which lit-up in different colors. The team were then able to detect real-time shape-shifting of the virus as the two beacons moved.

Using this technology, the researchers were able to analyze the movements of envelope proteins - gp120 and gp41, known as "trimers" - on the surface of the HIV virus.

Such proteins are important in allowing the HIV virus to infect human cells that carry CD4 receptor proteins - the proteins that assist HIV in binding to a cell. The researchers explain that these proteins "open up like a flower" when CD4 is present, revealing a gp41 subunit.

Smoking Finding ways to quit

The findings of the study, published in JAMA Internal Medicine, sug-gest that the disease burden of cigarette smoking in the US "remains immense," write the authors.

Cigarette smoking is acknowledged as the leading cause of prevent-able deaths in the US, with experts finding cigarette smoke capable of harming almost every organ and organ system within the body.

The Centers for Disease Control and Prevention (CDC) regularly publish their estimates of mortality attributable to smoking, along with its economic cost. However, the population disease burden tends to receive less focus, despite smoking's status as a leading cause for many major medical conditions.

In 2000, the CDC published estimates of smoking-attributable mor-tality and reported that 8.6 million individuals suffered from a total of 12.7 million medical conditions attributable to smoking.

"Most of these conditions were chronic bronchitis and emphysema, often classified as chronic obstructive pulmonary disease (COPD)," write the authors, "but these estimates and methods, to our knowledge, have not been subsequently updated or refined."
